Queen Elizabeth I founded the college in 1592 on the site of the former Augustinian Priory of All Hallows. The Book of Kells was produced around 800 AD, likely by monks at the monastery of Iona, Scotland, before being moved to Kells, County Meath, to escape Viking raids, and finally to Trinity in 1661.

The Long Room served as the visual inspiration for the Jedi Archives in the Star Wars film "Attack of the Clones," though the filmmakers did not officially film on-site. A long-standing student superstition claims that any student who walks beneath the Campanile (bell tower) while the bell tolls will fail their exams.
